The 'Challenge Enabled' option is the mechanism for determining the answerer of a call is human by asking that they 'press any number to accept this call'.
This setting can only be turned off for Ring Through call flows because it prevents multi number call flows from erroneously connecting the caller to an electronic answering service such as Google Voice.
If you're trouble shooting issues with your Ring Through call flows try switching between challenged and challenge-less options.
